【摘要】 以小鼠脾淋巴細(xì)胞為試驗(yàn)材料,將有絲分裂原刀豆蛋白A(ConA)或細(xì)菌脂多糖(LPS)與化療藥物5-氟尿嘧啶(5-Fu)聯(lián)合使用建立了體外免疫抑制模型,并比較了多糖樣品對(duì)5-Fu抑制淋巴細(xì)胞增殖的拮抗作用。結(jié)果表明,在供試樣品中,F1-1[龍須菜(Gracilaria lemaneiformis)中分離多糖]和BW[滬農(nóng)靈芝1號(hào)(Ganoderma lucidum)子實(shí)體中分離]對(duì)5-Fu產(chǎn)生的輕度抑制有完全拮抗作用,19號(hào)[滬農(nóng)靈芝1號(hào)(G.lucidum)子實(shí)體中分離]樣品刺激淋巴細(xì)胞增殖活性最好,但對(duì)5-Fu產(chǎn)生的抑制基本無拮抗作用,表明樣品刺激淋巴細(xì)胞增殖的活性與拮抗化療藥物抑制淋巴細(xì)胞增殖的活性無正相關(guān)性。

【Abstract】 National Research Center for Edible Fungi Biotechnology and Engineering; Key Laboratory of Applied Mycological Resources and Utilization,Ministry of Agriculture,the People's Republic of China; Key Laboratory of Agricultural Genetics and Breeding of Shanghai; Research Center of Natural Medicinal Resources,Institute of Edible Fungi,Shanghai Academy of Agricultural Sciences,Shanghai 201106,ChinaWe have developed an in vitro immunosuppression model based on the cell proliferation enhancing effects of the mitogens concanavalin A and lipopolysaccharide combined with the 5-fluorouracil (5-Fu)-induced inhibitory effect on mouse spleen lymphocyte proliferation. The model was used to compare the antagonistic effects of different polysaccharide preparations on 5-Fu-induced suppression of mouse spleen lymphocyte and mixed lymphocyte proliferation. Polysaccharide preparations F1-1 (from Gracilaria lemaneiformis) and BW (from Ganoderma lucidum) completely neutralized the mild inhibition of lymphocyt...更多e proliferation induced by 0.02 μg/mL 5-Fu. However,no positive correlation was found between the stimulating effect of a given polysaccharide on lymphocyte proliferation and the antagonistic effect of that polysaccharide on 5-Fu-induced suppression of lymphocyte proliferation. Thus,polysaccharide 19 (from G. lucidum) was most effective in stimulating lymphocyte proliferation but had no antagonistic effect on 5-Fu-induced suppression of lymphocyte proliferation.  還原
